This underlies important processes such as enzyme catalysis, binding of ligands at protein receptors and correct expression of genetic information at the level of protein. Even modest changes in structure (e.g. arising from point mutations or from interaction with environmental chemicals) can result in molecules with drastically altered biological activity. Several techniques have been presented elsewhere in this text which yield information on the approximate structure of biomolecules such as their quaternary structure, overall shape/mass and secondary structure (e.g. s 3, 4, 5, 7 and 10). These techniques often give us crucial information on the importance of particular structural domains for function and allow comparison of normal with aberrant molecules. However, in order to understand the detail of how biomolecules function, we need high-resolution struc tural information (i.e. at 2 A resolution). In practice, this information is particularly dif cult to obtain for the two most important classes of biomacromolecules (proteins and nucleic acids) and macromolecular complexes composed of mixtures of these such as ribosomes and viruses. Since proteins adopt a much wider range of tertiary (i.e. three-dimensional) structures than other biomacromolecules and frequently mediate the functioning of other classes of biopolymers, this chapter will concentrate in particular on protein structure. We will rst examine how proteins come to have a particular shape and then describe the two main experimental techniques which give us high-resolution insights into protein structure; multi-dimensional NMR and X-ray crystallography.
